VS-6766 is an orally available small molecule RAF/MEK clamp. VS-6766 is a dual RAF/MEK clamp that blocks both MEK kinase activity and the ability of RAF to phosphorylate MEK. Defactinib is an oral small molecule inhibitor of FAK and proline-rich tyrosine kinase (PYK2) that is being evaluated as a potential combination therapy for various solid tumors.
